What is Zonulin (Faecal), by immunoassay?
Zonulin was described by Fasano and colleagues as a physiological regulator of intestinal tight junctions, identified as pre-haptoglobin-2, the uncleaved precursor of haptoglobin-2. In the proposed mechanism it acts on the epithelial zonula occludens complex to reversibly loosen tight junctions and increase paracellular permeability, with gliadin and enteric bacteria described as triggers for its release. Tight junction regulation itself is well established as a genuine physiological process governing intestinal barrier function. What remains contested is whether the protein measured by commercial zonulin assays is the molecule described in that original work.
