What is Antinuclear Antibody Screen?
Antinuclear antibodies are immunoglobulins directed against nuclear constituents — chromatin, ribonucleoproteins, centromere proteins, or other nuclear antigens. Their presence reflects a breakdown in immune tolerance to self-antigens and is characteristic of systemic autoimmune rheumatic diseases, though low-titre ANAs occur in healthy individuals and rise in frequency with age. Indirect immunofluorescence on HEp-2 cells is the reference method because the substrate's dividing cells express a wide range of nuclear and cytoplasmic antigens, maximising sensitivity.
